Bacterial filtration efficiency (BFE) is, according to EN 14683 +AC, the main characteristic of medical face masks effectivity and quality. The method is based on the analysis of bacterial aerosol in Andersen cascade impactor (ACI) containing six stages simulating the different parts of the respiratory tract. The aerosol particles are separated on the base of their aerodynamic size and sediment on agar medium in Petri dishes filled in the impactor. The use of the glass Petri dishes is recommended for the correct particle sedimentation in ACI, but the most of laboratories prefer the disposable plastic dishes, actually. In our study, we evaluated the use of plastic dishes in Andersen impactor for the determination of BFE of medical face masks. The bacterium Staphylococcus aureus, used for BFE testing, allows only limited detection of submicron aerosol particles (on the sixth stage of the ACI). This limitation can be overcome by the use of bacteriophage aerosol and the method of viral filtration efficiency (VFE) testing. The bacteriophage Phi-X 174 with Escherichia coli host bacterium was chosen for VFE determination. The determined BFE and VFE values correlate well in the case of masks with high filtration efficiency (99 % and more). On the other side, lower values of VFE, comparing with BFE, were determined for masks with the filtration efficiency in the range of 95 – 98 %. There were detected higher numbers of plaque forming units (PFU) on the sixth stage of the Andersen impactor in the case of VFE determination in comparison with colony forming units (CFU) in the case of BFE determination. The VFE testing gives more exact information about the ability of tested mask material to filter viral particles carried by submicron aerosol particles (with the ability to penetrate the lungs). It seems appropriate to supplement the testing of face masks with the test of viral filtration efficiency.
The evolutionary pressure for life transitioning from extended periods of hypoxia to an increasingly oxygenated atmosphere initiated drastic selections for a variety of biochemical pathways supporting the robust life currently present on the planet. First, we discuss how fermentative glycolysis, a primitive metabolic pathway present at the emergence of life, is instrumental for the rapid growth of cancer, regenerating tissues, immune cells but also bacteria and viruses during infections. The ‘Warburg effect’, activated via Myc and HIF-1 respectively in response to growth factors and hypoxia, is an essential metabolic and energetic pathway which satisfies nutritional and energetic demands required for rapid genome replication. Second, we present the key role of lactic acid, the end-product of fermentative glycolysis able to move across cell membranes in both directions via monocarboxylate transporting proteins (i.e. MCT1/4) contributing to cell-pH homeostasis but also to the complex immune response via acidosis of the tumour microenvironment. Importantly lactate is recycled in multiple organs as a major metabolic precursor of gluconeogenesis and energy source protecting cells and animals from harsh nutritional or oxygen restrictions. Third, we revisit the Warburg effect via CRISPR-Cas9 disruption of glucose-6-phosphate isomerase (GPI-KO) or lactate dehydrogenases (LDHA/B-DKO) in two aggressive tumours (melanoma B16-F10, human Colorectal adenocarcinoma LS174T). Full suppression of lactic acid production reduces but does not suppress tumour growth due to reactivation of OXPHOS. In contrast, disruption of the lactic acid transporters MCT1/4 suppressed glycolysis, mTORC1, and tumour growth as a result of intracellular acidosis. Finally, we will briefly discuss the current clinical developments of an MCT1 specific drug AZ3965, and the recent progress for a specific in vivo MCT4 inhibitor, two drugs of very high potential for future clinical applications against cancers, bacterial and viral pathogens.
HIV-1 infection requires passage of the reverse transcribing viral core through the nuclear pore of the cell, a process that depends on activities of the viral capsid. Recent studies have challenged the idea that capsid disassembly is required for nuclear entry. Interactions with the nuclear pore complex are apparently necessary but not sufficient for nuclear entry, and the mechanism by which the viral core traverses the comparably sized nuclear pore is unknown. Here we show that the HIV 1 core is highly elastic and that this feature is linked to nuclear entry and infectivity. Using a novel atomic force microscopy-based approach, we found that purified wild type cores return to their normal conical morphology following a severe forced compression. Analysis of two HIV-1 mutants that exhibit impaired nuclear entry revealed that the mutant viral cores are brittle. A suppressor mutation reduced brittleness and rescue infectivity. Core elasticity was also reduced by treatment with Lenacapavir, a clinically relevant antiviral drug. Our results indicate that capsid elasticity is a fundamental property of the HIV-1 core that enables its passage through the nuclear pore complex, thereby facilitating infection. These results provide new insights into the mechanisms of HIV-1 nuclear entry and the antiviral mechanisms of HIV-1 capsid inhibitors.
